High-speed image reconstruction for optically sectioned, super-resolution structured illumination microscopy

چکیده
Super-resolution structured illumination microscopy (SR-SIM) is an outstanding method for visualizing the subcellular dynamics in living cells. To date, by using elaborately designed systems and algorithms, SR-SIM can achieve rapid, optically sectioned, SR observation with hundreds to thousands of time points. However, real-time still out reach most SIM setups as conventional algorithms image reconstruction involve a heavy computing burden. address this limitation, accelerated algorithm was developed implementing simplified workflow SR-SIM, termed joint space frequency reconstruction. This results 80-fold improvement speed relative widely used Wiener-SIM. Critically, increased processing does not come at expense spatial resolution or sectioning capability, demonstrated live imaging microtubule mitochondrial tubulation.

Localized plasmon assisted structured illumination microscopy for wide-field high-speed dispersion-independent super resolution imaging.
A new super resolution imaging method, i.e. Localized Plasmon assisted Structured Illumination Microscopy (LPSIM), is proposed. LPSIM uses an array of localized plasmonic antennas to provide dynamically tunable near-field excitations resulting in finely structured illumination patterns, independent of any propagating surface plasmon dispersion limitations. The illumination pattern feature sizes...
متن کاملMultifocus structured illumination microscopy for fast volumetric super-resolution imaging.
We here report for the first time the synergistic implementation of structured illumination microscopy (SIM) and multifocus microscopy (MFM). This imaging modality is designed to alleviate the problem of insufficient volumetric acquisition speed in super-resolution biological imaging. SIM is a wide-field super-resolution technique that allows imaging with visible light beyond the classical diff...
متن کاملSIMcheck: a Toolbox for Successful Super-resolution Structured Illumination Microscopy
Three-dimensional structured illumination microscopy (3D-SIM) is a versatile and accessible method for super-resolution fluorescence imaging, but generating high-quality data is challenging, particularly for non-specialist users. We present SIMcheck, a suite of ImageJ plugins enabling users to identify and avoid common problems with 3D-SIM data, and assess resolution and data quality through ob...
متن کاملOptically sectioned in vivo imaging with speckle illumination HiLo microscopy.
We present a simple wide-field imaging technique, called HiLo microscopy, that is capable of producing optically sectioned images in real time, comparable in quality to confocal laser scanning microscopy. The technique is based on the fusion of two raw images, one acquired with speckle illumination and another with standard uniform illumination. The fusion can be numerically adjusted, using a s...
